Novo Nordisk's investigational drug zenagamtide has shown an average weight loss of up to 14.6% in individuals with type-2 diabetes. According to the data from the positive phase 2 trial presented at the 2026 Scientific Sessions of the American Diabetes Association (ADA) in New Orleans, LA, zenagamtide is the first drug of its kind to combine two separate biological keys that control metabolism into one single molecule: a GLP-1 receptor agonist and an amylin receptor agonist. Unlike older weight loss medications like semaglutide (Wegovy/Ozempic), which target only one hormone receptor, GLP-1, zenagamtide is a unimolecular peptide agonist that targets two distinct metabolic pathways simultaneously: GLP-1 Receptor Agonist: Mimics the GLP-1 hormone to stimulate insulin secretion, lower blood sugar, and target brain regions to reduce hunger. Amylin Receptor Agonist: Mimics amylin, a hormone co-secreted with insulin that suppresses appetite, slows down food transit, and enhances feelings of fullness. By combining these two mechanisms into a single molecule, it maximises appetite suppression and glucose regulation.
